摘要
目的建立五味子浸膏中五味子醇甲和五味子乙素含量测定方法。方法高效液相色谱法,采用LichrosphereC18柱(4.6mm×250mm,5μm);流动相为水(A)-甲醇(B)梯度洗脱(0~15min,60—70%B;15—25min,70%~80%B;25~35min,80~75%B;35~45min,75%B)。流速1.0ml·min^-1,柱温30℃,检测波长254nm。结果五味子醇甲和五味子乙素分别在0.1824—1.6416μg(r=1.0003)和0.1896—1.7064μg(r=0.9999)范围内线性关系良好,平均加样回收率分别为98.02%和98.42%。结论该法简便、准确、专属性强,可用于五味子浸膏的质量控制。
Objective To establish a method for the determination of schisandrin and schisandrin B in extract from fruits of Schisandra chinensis. Methods An HPLC method was set up with Lichrosphere C18 (4.6 mm ×250 mm,5μm) column. The Mobile phase was H2O(A) and MeOH(B). Gradient elution was with 60 -70% B from 0 - 15 min, 70% - 80% B from 15 - 25 min, 80% -75% B from 25 -35 min, and 75% B from 35 - 45 min. The flow rate was 1.0 ml· min^-1, the column temperature was 30℃ and detection wavelength was 254 nm. Results The linear ranges of schizandrin and schisandrin B were within 0. 182 4 - 1. 641 6μg(r = 1. 000 3) and 0. 189 6 - 1. 706 4 μg(r = 0 . 999 9), the average recoveries were 98.02 % % and 98.42 % respectively. Conclusion This method is easy, sensitive, specific and accurate for the determination of schisandrin and schisandrin B in extract from fruits of Schisandra chinensis.
