摘要
利用日本DDBJ数据库电子克隆了条斑紫菜的6-磷酸海藻糖合成酶基因(pytps),得到全长cDNA序列2727bp;经过ORFfinder分析,获得了相应蛋白质的全长序列908Aa,分子量约为101.8kD。将条斑紫菜的6-磷酸海藻糖合成酶与多种模式生物大肠杆菌、裂殖酵母、拟南芥、水稻、秀丽隐杆线虫、黑腹果蝇的同源蛋白进行序列比对得到了聚类分析图表明它们之间具有一定的进化相关性功能结构域预测分析显示PyTPS拥有两个功能结构域Glyco_transf_20 domain和Trehalose_PPase domain,这对于进一步分析蛋白质结构与功能的关系将有很大的启示。
TPS encoded by the sane tps is one of the key enzymes involved in trehalose biosynthesis.In this work, all ESTs related to PyTPS (TPS for Porphyra yezoensis ) were collected from the database of DDBJ. Then they were joined together and the total cDNA sequence was obtained with the length of 2 727bp. And the corresponding protein was 908 amino acids and with the lnoleeuiar weight 101.8kD.Sequence holnology analysis was carried between PyTPS and TPSs in other model organisms ( Escherichia coil, Schizosoccharomyces point,era, Arbidopsis thaliana, Oryza sots, Caenorhobditis elegans, Drosophila melanogaster,all the sequences were from NCBI database. ). The re.sults showed that PyTPS has halnologous relationship with other organisms. The functional domains of PyTPS were predicted by using the NCBI CDD program. And two functional domains (Glyeo.transf 20 domain and Trehalose PPase domain) were found in the protein.
