丹酚酸B对局灶性脑缺血/再灌注大鼠血清超氧化物歧化酶活力和丙二醛含量的影响 被引量:21

Influence of Salvianolic acid B on serum SOD activities and MDA concentrations in rats with focal cerebral ischemia-reperfusion

摘要 [目的]观察丹酚酸B对大鼠局灶性脑缺血/再灌注损伤时血清超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量的影响,探讨该中药单体防治缺血性脑血管疾病的作用机制。[方法]健康雄性Wistar大鼠24只,随机分成3组:假手术组、模型组、丹酚酸B组,每组8只。丹酚酸B组预先给药,1次/d,连续7d。末次给药后1h除假手术组外其他两组大鼠线栓法制备大脑中动脉局灶性缺血再灌注模型,缺血2h,再灌注24h。实验结束时测定各组大鼠神经功能缺失评分及血清总SOD活力和MDA含量。[结果]模型组神经功能缺失评分升高,血清总SOD活力下降,MDA含量增加;丹酚酸B组能降低神经功能缺失分值,提高血清总SOD活力并减少MDA含量。[结论]丹酚酸B能改善缺血/再灌注损伤大鼠的神经功能,减少抗氧化酶的消耗,促进自由基的清除,并减轻脂质过氧化反应。 [Objective] To study the effects of salvianolic acid B on the serum superoxide dismutase (SOD) activities and malondialdehyde (MDA) concentrations in rats with focal cerebral ischemia-reperfusion injury and analyze the mechanism of action of salvianolic acid B in preventing and treating ischemic cerebrovascular disease. [Methods] Twenty-four healthy male Wistar rats were randomly divided into three groups with eight rats in each group: sham operation group, model group, salvianolic acid B groups. The latter group was infused with corresponding drugs once a day for seven days. Then the intraluminal suture technique was used to establish the models of middle cerebral artery occlusion (MCAO) for 2 h of occlusion and 24 h of reperfusion in rats except sham-operation group. At the end of reperfusion, the extent of neurological deficits was evaluated by Longa's model. And the serum SOD activities and MDA concentrations were monitored after the focal cerebral ischemia-reperfusion. [Results] In model group, the score of neurological deficits was raised, serum SOD activities were decreased and serum MDA concentrations were increased. Salvianolic acid B could reduce the score of neurological deficits, increase serum SOD activities and decrease MDA concentrations. [Conclusions] Salvianolic acid B can improve neurological function of rats with focal cerebral ischemia-reperfusion injury, reduce the consumption of antioxidase, promote the elimination of free radical and relieve lipid peroxidation reaction.
