绵羊体细胞克隆胚胎的异常DNA甲基化模式 被引量:5

摘要 以体外受精胚胎作为对照,利用抗5-甲基胞嘧啶抗体免疫荧光法对绵羊体细胞克隆胚胎的基因组甲基化模式进行了分析.实验结果表明:(ⅰ)在着床前发育过程中,体细胞克隆胚胎呈现出与体外受精胚胎类似的去甲基化趋势,即在8-细胞期甲基化水平降到最低点,紧接着在桑椹胚时又升高,但是克隆胚胎的甲基化水平明显高于同一时期的体外受精胚胎,特别在8-细胞期及其以后时期;(ⅱ)克隆囊胚的甲基化模式与体外受精囊胚不同,克隆囊胚的内细胞团(inner cell mass,ICM)和滋养层(trophectoderm cells,TE)甲基化水平相当,而体外受精囊胚的内细胞甲基化水平低于滋养层.研究结果表明,与体外受精胚胎相比,克隆胚胎的DNA甲基化重编程存在异常,它可能是导致克隆胚胎发育失败的原因之一.
