衰老大鼠淋巴细胞磷酸化p65,IκBα,IκBε表达特点及淫羊藿总黄酮的干预研究 被引量:9

摘要 目的:研究衰老状态下脾淋巴细胞磷酸化p65,IκBα,IκBε的表达特点,并探讨淫羊藿总黄酮(EF)的干预作用。方法:将SD大鼠随机分为4月龄(4 m)、27月龄(27 m)、27 m+EF组,其中检测IκBε,IκBα时,对27 m及27 m+EF组均用NF-κB信号通路抑制剂二硫代氨基甲酸吡咯烷(PDTC),进行NF-κB阻断(即27 m PDTC及27m PDTC+EF组)。以SD大鼠脾脏分离的淋巴细胞为研究对象,采用Western-blot方法,分别比较检测各组淋巴细胞磷酸化p65,IκBε,IκBα的差异表达。结果:衰老状态下,RelA(p65)、磷酸化p65,IκBα,IκBε的表达与青年组对比,均明显降低,对27 m组进行PDTC阻断后则愈加明显。而加用EF干预后,老年各组的以上指标表达均可明显上调。结论:衰老大鼠淋巴细胞中p65,IκBα,IκBε磷酸化蛋白表达明显不足,EF能够显著上调衰老状态下以上蛋白的表达。 Objective: To study the characteristics of protein phosphorylation of p65,IκBα and IκBε in lymphocytes of rats in the progress of aging and the the interventional effect of Epimedium flavonoids(EF).Method: We chose the lymphocytes derived from SD rat spleen.We divided the SD rats into five groups i.e.4 months(4 m),27 months(27 m),the 27 m EF teeated group(27 m+EF),and we also observed the whole interventional effect of PDTC(a NF-κB inhibitor) on old rat groups(27 m PDTC,27 m PDTC+EF) when IκBε,IκBα were detected.Through the western-blotting analyses,we studied the entire characteristics and distinctions of phosphorylation expression in molecules related to NF-κB signal transduction pathway p65, IκBε and IκBα in lymphocytes across the age spectrum of rats in aging.We also observed the whole interventional effect of EF on lymphocytes of old rats. Result: With the increasing of age,the mean level of phosphorylation expressions of p65,IκBα and IκBε in rat spleen lymphocytes decreased obviously,When inhibited NF-κB by PDTC,there was decreased evidently,while PDTC+EF can active NF-κB family and the above molecules were increased to a certain extent.Conclusion: The phosphorylation expressions of p65,IκBα and IκBε in rat spleen lymphocytes were not enouphe,EF have a strong effect to upregulated the expression of them during aging.
