摘要
目的观察糖神胶囊对糖尿病大鼠肾损害的保护作用,并探讨黄芪在糖神胶囊中的最佳剂量。方法建立链脲佐菌素(STZ)诱导糖尿病大鼠模型,分正常对照组(NC组)、糖尿病组(DM组)、高剂量黄芪糖神胶囊治疗组(HDT组)、低剂量黄芪糖神胶囊治疗组(LDT组)。用药8周后检测大鼠空腹血糖(FBG)、血清果糖胺(FIM)、血肌酐(Scr)、肌酐清除率(Ccr)、尿微量白蛋白(UAlb)、红细胞中山梨醇含量(RBCS)、肾重/体重等。行大鼠肾脏PAS染色测定肾小球平均截面积(MGA)及体积(MGV);免疫组化染色观察细胞间黏附分子1(ICAM-1)的表达。结果①HDT、LDT组较DM组Scr下降、Ccr升高(P<0.05),UAlb、RBCSF、BG、FIM、肾重/体重、MGA、MGV均下降(P<0.05);②HDT、LDT组均可降低ICAM-1的表达。上述指标以HDT组改善最明显。结论糖神胶囊能有效减轻糖尿病大鼠肾损害。黄芪在糖神胶囊中的最佳剂量为120 g/料。
Objective To observe the protective effects of the Tangshen capsule on nephropathy damages in diabetic rats and explore the optimal dosage of Astragalns membranacens in the drug. Methods Diabetic rots were induced by streptozotocin （STZ） and were divided into the diabetic model group（DM）, the high-dose treatment group （HDT group）, and the low-dose treatment group （LDT group）. After an 8-week treatment, serum creatinine （Scr）, urine albumin （UAIb）, sorbitol content in red blood cell （RBCS）, fasting blood glucose （FBG）, fructosamine （FIM）, triglyceride （TG）, cholesterol （CHO） and the kidney hypertrophy index were determined. The ultrastructure of the rats＇ kidneys was observed after PAS staining. The expression of intercellar adhesion molecule-1 （ICAM-1 ） was determined by the immtmohistochemical technique. Results ① Compared with the DM group, each treatment group had a higher weight, lower Scr, UAIb, RBCS, FBG, FIM and kidney hypertrophy index（ P 〈 0.05）; ② Compared with the NC group, the expression of ICAM-1 was up-regulated in the other groups. But in treatment groups, the above items were ameliorated （ P 〈 0.05） and they did the best in the HI）T group. Conclusion The Tangshen capsule can offer protection against kidney damage of diabetic rats and the optimal dosage of Astragalns Membranaceus is 120 g per powder.
