摘要
在pH=9.9的Brittion—Robinson介质中,两性离子染料五号专利兰与核酸以及阳离子表面活性剂十六烷基三甲基溴化铵作用形成三元复合物,使得345nm处的共振光得到加强.采用共振光散射(RLS)技术研究了五号专利兰一十六烷基三甲基溴化铵一核酸三元体系的紫外一可见吸收光谱特征、共振散射光谱特征、共振散射光谱的影响因素和最佳反应条件.在最佳条件下,体系的ΔIRLs与鱼精子脱氧核糖核酸(fsDNA)、小牛胸腺脱氧核糖核酸(ctDNA)、酵母核糖核酸(yRNA)分别在0~2.0mg/L、0-1.6mg/L和0~1.5mg/L的浓度范围内呈线性关系,检测限分别为10.2mg/L、15.4mg/L和14.6mg/L.该方法可应用于合成样品中核酸的测定.
The interaction of Patent Blue V ( PBV ) with DNA in the presence of cetyhrimethyl - ammonium bromide has formed a ternary complex of PBV - CTMAB - DNA in Brittion - Robinson buffer of pH = 9.9, which result in significant enhancement of resonance light scattering ( RLS ) intensity. The spectral characteristics of UV - Vis absorption and RLS, the affecting factors and optimum conditions of the reaction have been investigated. Under the optimum conditions, the enhanced RLS intensity is proportional to the concentration of nucleic acids, over the ranges of 0 - 2.0 mg/L for fish sperm DNA ( fsDNA ), 0 - 1.6 mg/L for calf thymus DNA ( ctDNA ) and 0 - 1.5 mg/L for yeast RNA ( yRNA ) , with the limits of determination of 10.2 mg/L for fsDNA, 15.4 mg/L for ctDNA and 14.6 mg/L for y RNA,respectively. This method has been applied to determination of DNA in synthetic samples.
