摘要
[目的]为获得纤维素分解菌,以研究其在有机物质资源的利用上的应用。[方法]通过固体培养初筛和摇瓶发酵复筛从腐烂的含菌秸秆、腐叶、朽木和土壤中筛选出纤维素酶活较高的菌株,并对它们对不同碳源的利用进行了初步研究。[结果]通过初筛和复筛获得了内切酶活力和滤纸酶活力均较高的3株细菌和4株真菌。将7个菌株分别接种到不同碳源的液体培养基中,经3d培养后测定其CMC酶活,可发现不同菌株对同一碳源产生的酶活不同,同一菌株对不同来源纤维素的利用能力也不同。[结论]测定各菌株在不同纤维素碳源中的纤维素酶活力并将其应用于相应行业,这在生产上具有重大意义。
[Objective] The research aimed to obtain cellulose-decomposing microorganisms and study their applications in making use of organic matter resources.[Method] Strains with higher cellulase activity were screened out from rotten straw,leaves,wood and soil with bacterium through primary screening of solid culture and secondary screening of shaking-flask fermentation and their utilization on different carbon sources was preliminarily studied.[Result] 3 bacterial strains and 4 fungal strains with higher endonuclease activity and filter paper enzyme activity were obtained through primary screening and secondary screening.These 7 strains were respectively inoculated on liquid medium with different carbon sources to determine their CMC activity after 3 d culture.It was found that the enzyme activities of different strains to the same carbon source were different and the utilization capabilities of the same strain to different source cellulose were different.[Conclusion] It was of great significance in the production to determine the cellulase activity of each strain in different cellulose carbon resources and apply them in the corresponding industries.
