摘要
目的探讨洛伐他汀对急性冠脉综合征(ACS)患者外周血单核细胞ABCA1蛋白表达的影响。方法选取急性冠脉综合征患者21例,密度梯度法分离外周血单核细胞。洛伐他汀分别在同一浓度(10μmol/L)、不同时间(0、6、12、24h),同一时间(24h)、不同浓度(0、1、10、100μmol/L)条件下与单核细胞在细胞培养箱中孵育,孵育结束后用流式细胞技术检测外周血单核细胞ABCA1蛋白。结果与对照组比,洛伐他汀作用单核细胞后,ABCA1蛋白表达无明显变化(P>0.05)。结论洛伐他汀不能诱导ABCA1蛋白表达增加,其升高高密度脂蛋白(HDL)的机制可能与ABCA1蛋白无关。
Objective To investigate the effect of lovastatin on ABCA1 （ATP-binding cassette trans- porter- 1 ） of peripheral blood mononuclear cell in patients with acute coronary syndrome. Methods 21 patients were enrolled in the study. Peripheral blood monocytes （PBMC）were isolated by gradient eentrifugation. Monocytes incubated with 10 μmol/L lovastatin for 0,6,12,24 h,and incubated with lovastatin at different concentration （0,1.0, 10,100 μmol/L）for 24 h. The level of protein ABCA1 were subjected to flow cytometric detection. Results Compared with control group, ABCA1 protein expression had no significantly difference （P〉0.05）.Conclusion Lovastatin could not increase the expression of protein ABCA1. It implied that statin increasing HDL was not re- lated with ABCA1.
