咳嗽变异性哮喘豚鼠模型的构建 被引量:35

摘要 目的尝试制备一种咳嗽变异性哮喘(CVA)的豚鼠模型。方法30只雄性豚鼠饲养1周后,随机分为健康正常组和模型组,每组15只。模型组第1天肌注4%OVA溶液0.5ml,同时腹腔注射2%Al(OH)30.2ml,14d后,模型组用1%OVA溶液雾化后攻击,隔天1次,每次15s,共7次。第29天,两组豚鼠用10-4mol/L辣椒素溶液引咳。结果通过对咳嗽次数、肺病理切片、肺泡灌洗液、外周血等指标的观测,发现模型组的咳嗽次数多于健康正常组;与健康正常组比较,模型组的病理损伤明显;从肺泡灌洗液和血内嗜酸性粒细胞计数及白细胞计数上看,模型组与健康正常组两组存在统计学差异,前者较后者计数均有升高。结论这种改进后方法能成功地建立咳嗽变异性哮喘模型。 Objective To establish a cough variant asthma (CVA) model in guinea pig. Methods Thirty male guinea pigs were randomly divided into two groups: normal group(n= 15) and model group(n = 15). The guinea pigs in model group were sensitized with intramuscular injection of 4% ovalbumin solution 0.5 ml and intraperitoneal injection of AI(OH)3 solution 0.2 ml at day 1. After 14 d, the guinea pigs were challenged with aerosolized 1% ovalbumin(OVA) solution for 15 s every other day . At day 29, all guinea pigs were challenged with aerosolized 10^-4 mol/L capsaicin solution for violent cough and corresponding pathological alteration . Results Compared with normal animals , theguineapigsinmodelgroup had higher cough frequency [ ( 7.82 ± 2.44 ) vs (3.67±3.16),P=0.004], higher white blood cell(WBC) counts[(2.44±0.91) × 109/L vs (1.66±0.56) × 10^9/L, P=0.034] and eosinophils(EOS) counts[ (0.12 ± 0.07) × 109/L vs (0.05 ± 0.03) × 10^9/L, P = 0. 002 ] in bronchoalveolar lavage, higher WBC counts [(11.09±2.26)× 109/L vs (8.30± 1.90) × 109/L,P = 0.006] and IK)S counts[(0.12±0.06) × 10^9/L vs(0.08± 0.03) × 10^9/L, P = 0. 014] in blood. There was significant pathological impairment in model group. Conclusion CVA model can be established successfully by this method.
