摘要
对9a生邓恩桉优株树干基部环割促萌,以萌芽条的茎段为外植体,进行始芽诱导、不定芽增殖以及再生植株的培养研究。结果显示:以0.1%HgCl2处理外植体8~10min时无菌活外植体得率较高,达到50%~60%;适宜茎段离体培养的基本培养基为改良B5,即B5+NH4NO360mg/L;细胞分裂素与生长素组合6-BA0.18mg/L+NAA0.14mg/L有利于不定芽的增殖和生长,培养25d时不定芽增殖系数3,且生长健壮;单芽在1/2B5+IBA1.2mg/L+NAA1.6mg/L上生根率为73.3%。
The superior trees of nine-year-old Eucalypts dunnii were selected and treated with girdling to promote sprouting. The stem segments from shoots of eucalypts were used as explants to carry out the shoots induction, adventitious shoot proliferation and plant regeneration. The results showed that the explant survival rate was higher and reached to 50% - 60% when the explants were treated with 0. 1% HgCl2 for 8- 10 min. The optimal medium for stem segments in vitro culture was the modified B5( B5 +60 mg/L NH4NO3 ). The 6-BA 0. 18 mg/L+NAA 0. 14 mg/L added was propitious to proliferation and growth of adventitious shoot and the proliferation coefficient reached to 3 after 25 days. The adventitious shoots grew well. The rooting rate of a single shoot was 73.3% in the medium of IBA 1.2 mg/L+NAA 1.6 mg/L+1/2B5.
