摘要
以菘蓝为材料,用碘-碘化钾(I-KI)染色法、氯化三苯四氮唑(TTC)染色法、亚甲基蓝染色法和琼脂培养法对花粉生活力进行测定。结果表明:用I-KI染色法和TTC染色法测定花粉生活力时都不理想,有生活力和没有生活力的花粉在光学显微镜下不易区分,亚甲基蓝染色法效果明显,但结果偏高,添加20 mg/L蔗糖和3 mg/L硼酸的琼脂培养基花粉萌发率达到90.3%以上,高于其他组分培养基。
Pollen viability of indigotica is surveyed and evaluated by I - KI stained method, TYC stained method , methylene blue stained method and agar culture medium. The result indicate that I - KI stained method, TYC stained method are all unsuitable because the viable pollen can not be distinguished with exanimate pol- len ; the effects of methylene blue stained method is remarkable but the viability index is more than actual index ; pollen germination rate in agar culture medium contained 20 mg/L cane sugar and 3mg/L boric acid is more than 90.3 percents and its germination rate is higher than other agar culture medium.
