血液中白细胞与红细胞更新速度不同,它们所含微量元素的量可分别反映机体即时及长期元素的水平,它与某些疾病或慢性金属职业中毒有关。因此,它是医学中新的检测对象,具有实际意义。但是,白细胞数仅为红细胞数的1‰,其中微量元素含量更少,故不易测定,其关键是分离白细胞,以提高火焰原子吸收法的检测能力。本文以淋巴细胞分离液重力离心法分出白细胞与红细胞,方法较简便结果可靠。
Erythocytes and leucocytes of blood in separative liquid of lymphocytes were separated by using gradient centrifugation. Leucocytes on the surface and fixed to certain volume, in which the contents of Zn, Cu, Mn, and Mg were determined with atomic absorption spectrometry. Some steps were adopted to increase the accuracy. The over all recovery of the method was 86.96-104.5%, which is suited to the need of the method.
