中低温度热疗诱导骨肉瘤细胞株凋亡 被引量:2

Apoptosis of osteosarcoma cells induced by middle-low temperature thermotherapy

摘要 目的:研究加热诱导成骨肉瘤细胞株(OS-9901)凋亡,为中低温度热疗辅助成骨肉瘤综合治疗提供理论依据.方法:成骨肉瘤OS-9901细胞株分别经不同温度的热疗作用1h,用TUNEL法染色及计数、透射电子显微镜观察其对骨肉瘤细胞株的诱导凋亡作用.同时通过免疫组织化学染色,观察43℃1h热疗对成骨肉瘤细胞株OS-9901的Bcl-2蛋白表达的影响.结果:43℃作用1h的成骨肉瘤细胞株继续培养6h后出现大量异常细胞,透射电镜观察可见有典型的细胞凋亡特征性形态改变.TUNEL法染色后通过计数可见有42%的细胞凋亡比例.免疫组织化学显示,43℃作用1h的成骨肉瘤细胞株6h后Bcl-2蛋白的表达明显减少.结论:43℃作用1h可以诱导成骨肉瘤细胞株OS-9901凋亡.加热后成骨肉瘤细胞Bcl-2表达减少,从而进一步诱导发生细胞凋亡,为热疗诱导肿瘤细胞凋亡的机制提供了部分理论依据. AIM: To observe the apoptosis of the osteosarcoma cells ( OS-9901 ) induced by hyperthermia and to explore the possible mechanism for mild temperature treatment of osteosarcoma. METHODS: The osteosarcoma 0S-9901 cells were treated with different temperatures for 1 h. The cell apoptosis was analyzed with electron microscope, TUNEL staining and counting. The Bcl- 2 protein expression in the OS-9901 cells was also observed before and after hyperthermia (43℃, 1 h) using the immunohistochemical technology. RESULTS: A great number of abnormal osteosarcoma cells were observed at 6 h after 43℃ treatment for 1 h. Transmission electron microscopy demonstrated typical apoptosis morphological features of the cells, including size reduction, chromatin condensation, and apoptotic bodies appearance. The highest apoptosis rate reached 42% analyzed by TUNEL staining. The immunohistochemistry showed that the expression of Bcl-2 decreased after treatment with 43℃ for 1 h. CONCLUSION: 43℃ treatment for 1 h induces apoptosis of osteosarcoma cells. Hyperthermia decreases the Bcl-2 protein expression in the osteosarcoma cells, so as to induce the apoptosis of the cells . This study provides some theoretical basis for the mechanism underlying the apoptosis of tumor cells induces by thermotherapy.
