摘要
通过对三种枯草芽孢杆菌的比较分析,以其中酶活力最高的一株作为出发菌株,用15W紫外灯于30cm处照射120s,获得菌株UV-12-3,酶活力为4238.6u/mL。通过正交实验,确定其最适水解条件为:pH6.5、温度32℃、水解42h、接种量为8%。在此条件下水解度可达到46.7%。
One strain which was fit for the hydrolysis of defatted soybean, was screened out from three Bacillus subtilis strains. Treatment with ultraviolet radiation of 15W was made for 120s and with a distance of 30cm. And then UV- 12 -3 with high production of protease was selected. The protease vigor was 4238.6u/mL. Through the orthogonal test, the optimum conditions of hydrolysis were ascertained: 8% inoculating amount, pH 6.5 , temperature 3213, and the time of fermentation 42h. Under this condition the hydrolysis degree can reach 46.7% .
