光损伤RPE细胞中EPO、EPOR的表达及意义

摘要 目的检测光化学损伤后视网膜色素上皮(RPE)细胞中促红细胞生成素(EPO)及其受体(EPOR)的表达,并探讨其意义。方法成人ARPE-19细胞株传代培养2-5代,建立光损伤模型,分别于光照后6、12和24h终止培养,采用免疫组化SP法和W estern b lot免疫印迹法检测EPO和EPOR在RPE细胞中的表达。结果EPO、EPOR表达均于培养终止后12 h达到高峰,24 h开始下降。结论EPO及其受体EPOR在光损伤RPE细胞中表达升高;这可能是光化学损伤时RPE细胞的自我保护性机制之一。 [ Objective ] To assess the expression and significance of erythropoietin (EPO) and erythropoietin receptor (EPOR) in human retinal pigment epithelial (RPE) cells exposed to light. [ Methods]The 2 -5 generation cultured human RPE cells were exposed to light of (2000 ± 500) lux energy for 12 h to establish light induced injury model, and the culture was finished after 6,12 and 24 h respectively. Immunocytochemical staining and Western blot were used to determine the expression of EPO and EPOR in human RPE cells. [Results]In the 12 h treated group, the expression of EPO and EPOR reached the peak, and began to decrease after 24 h. [ Conclusion ] Expression of EPO and EPOR in human RPE cells can be promoted by light - induced injury. It may be one of the protective mechanisms of RPE cells in injured by light.
