摘要
目的:建立氧氟沙星(OFL)的ELISA快速检测方法。方法:在制备抗OFL单克隆抗体基础上,采用间接竞争酶联免疫吸附方法(ciELISA)检测OFL的含量。结果:筛选得到特异性分泌抗OFL的单克隆抗体细胞株3G3和3D9,应用3G3所制备腹水建立的ciELISA工作曲线的线性范围为0.5-128 ng/mL(r^2=0.985 8),最低检出浓度为0.5 ng/mL,半数抑制浓度IC50为7.0 ng/mL,样品加样回收率为84%-120%,批内变异系数和批间变异系数均小于15%。结论:本文建立的氧氟沙星ELISA检测法可满足OFL残留检测的需要。
Aim. To develop ELISA for the assay of ofloxacin. Methods: Ofloxacin was determined by indirect competitive inhibition enzyme-linked immunosorbent assay (ciELISA) on the basis of monoclonal antibody against ofloxacin. Results:Two hybridoma lines 3G3 and 3D9 were isolated.The linear range of the ciELISA using 3G3 had a linear range of 0.5 - 128 ng/mL ( r^2 = 0. 985 8). The low of detection (LOD) of the assay was 0.5 ng/mL, and IC50 was 7.0 ng/mL. The recovery of ofloxacin spiked in samples was among 84% - 120%. Both the inter-assay and intraassay coefficient of variation (CV) was less than 15%. Conclusion: The established method is rapid, sensitive and specificity and could meet the requirements in the determination of ofloxaein residue.
