摘要
采用回交法,将抗除草剂转基因小麦bar基因导入皖麦48和扬麦158中。经苗期抗性鉴定及GS酶活性、叶绿素含量、PAT酶活性测定,结果证实了抗除草剂bar基因已经整合到小麦基因组中且能够正常表达。回交后代与对照在主要农艺性状上无明显差异,表明通过回交将bar基因转育到其它品种,是获得新的抗除草剂小麦品种的一条简便有效的途径。
A herbicide resistant gene bar was transferred into two cultivated wheat Wanmai 48 and Yangmai 158 by backcrossing. The test of herbicide-resistance, chlorophyll content, glutamine synthetase (GS) activity and phosphinthricin acetyltransferase(PAT) activity indicated that the backcross generations B2F1 showed positive reaction of the bar gene. It suggested that the foreign gene bar had been transferred into and was expressed in two cultivated wheat. Most of agronomic characters of the backcross generations B2 F1 were similar to their recurrent parents. The results confirmed that transfering bar gene into other variety by backcrossing is a simple and effective approach to get a new wheat variety resistant to herbicide PPT.
