IL-12基因修饰瘤苗治疗小鼠眼内移植性黑色素瘤疗效观察 被引量:2

Therapeutic effect of IL-12 gene modified tumor cell vaccine on transplanted intraocular melanoma in mice

摘要 目的观察白细胞介素12(IL-12)基因修饰瘤苗对移植性黑色素瘤的疗效。方法将荷瘤模型鼠分为PBS治疗组,野生型肿瘤细胞疫苗(WTV)治疗组,IL-12基因修饰肿瘤细胞疫苗(GTV)治疗组。模型建立后的第3d、10d分别在各组鼠结膜下注射30μlPBS、30μl(3.0×106个细胞)灭活的WTV及GTV,观察角膜溃破时间、淋巴结和肺的转移率及生存时间。结果PBS组及WTV组角膜溃破时间早于GTV组,前两组间差异无统计学意义;模型建立后的第18d,GTV组淋巴结转移瘤发生率低于PBS组及WTV组,后两组间差异无统计学意义;模型建立后第28d的各组间小鼠肺转移瘤发生率及各组间小鼠生存时间均无统计学意义。结论IL-12基因修饰瘤苗对实验模型鼠荷瘤早期起到了一定的抗肿瘤效应。 Objective Many researches has clarified the therapeutic effect of interleukin-12 (IL-12) to many types of tumors ;while the therapeutic effect of IL-12 gene modified tumor cell vaccine to tumor is becoming a hot topic. The effectiveness of IL-12 gene modified tumor cell vaccine on transplanted intraoeular melanoma was observed in this study. Methods Twentyfour C57BL/6 model mice bearing tumor were divided into PBS treatment group,wild type tumor cell vaccine (WTV) treatment group and gene modified tumor cell vaccine (GTV) treatment group. 30 μl PBS,30 μl (3.0 × 10^6 cells)inactivated WTV and GTV were respectively injected into subeonjunetiva of mice bearing tumor 3 days and 10 days after modeled. The time of cornea diabrosis,metastases into lymph nodes and lung as well as survival time were observed. Results The time of cornea diabrosis in PBS and WTV groups was earlier than that in GTV group( P 〈 0.05 ) , hut there was no statistical difference between PBS group and WTV group. Metastases in ipsilateral cervical lymph nodes of eyes transplanted tumor cells were developed in mice of various group 18 days after modeled. There was statistical difference in metastatic incidence between PBS group, WTV group and GTV group( P 〈 0.05 ) ,but no statistical difference between PBS group and WTV group. Metastases in lungs were developed in mice of every group 28 days after modeled. There was no statistical difference in lung metastatic incidence and survival time between every group. Conclusion IL-12 gene modified tumor cell vaccine can elicit anti-tumor effect in some extent against the intraocular melanoma in mice, especially in early stage of tumor.
