摘要
目的检测重组日本血吸虫谷胱甘肽-S-转移酶可生物降解微球疫苗免疫小鼠的血清学指标动态变化。方法利用已构建的表达质粒pET28a(+)-SiGST在E.coli BL21内表达的日本血吸虫谷胱甘肽-S-转移酶融合蛋白,经纯化后的蛋白质以聚乳酸/乙醇酸(PLGA75/25)为包裹材料,制备rSjGST可生物降解微球。按一定剂量,将其单针皮下注射免疫6周龄BALB/c雌性小鼠,分别于免疫后6、9、12、15、18、21周眼眶采血,通过检测血清中特异性IgG、IL-2及IFN-γ的动态水平,观察其免疫效果。结果与对照组相比,各免疫组血清特异性IgG水平在6、9周时差异均有统计学意义(P均〈0.01),弗氏完全佐剂(FCA)组、铝佐剂组均在免疫后9周达峰值,后逐渐下降,而微球组在21周时仍呈上升趋势(P〈0.01);微球组、FCA组及铝佐剂组IL-2、IFN-γ水平在6、9周时差异均有统计学意义(P分别〈0.05、0.01、0.01),但微球组在12~21周时仍呈上升趋势(P〈0.01),FCA组、铝佐剂组则在12周后逐渐下降。微球组分别与FCA组和铝佐剂组间比较,微球组血清特异性IgG水平在12周后、IL-2在18周后及IFN-γ在15周后差异均有统计学意义(P均〈0.01)。结论重组日本血吸虫谷胱甘肽-S-转移酶可生物降解微球免疫小鼠,具有明显长效和高效作用。
Objective To detect the dynamic changes of antibody and eytokines in the serum induced by single vaccine delivery system (SVDS) of biodegradable mierospheres of recombinant glutathione-S-transferase from Schistosoma japonicum (rSiGST) in mice. Methods By using the constructed expression plasmid pET28a(+)-SjGST, rSjGST was expressed by E. coli BL21. After purified through the column of resin,the biodegradable mierospheres of rSjGST were prepared with PLGA (Poly laetie-eo-glyeolie acid, PLGA 75/25). Six-week BALB/c female mice were immunized subcutaneously with a single vaccine, and the blood was collected from eyes on the 6th, 9th, 12th, 15 th, 18th, 21th week respectively. The dynamic levels of serum specific antibody IgG, IL-2 and IFN-γ were detected. Results Compared with the control group, there was a significant difference of serum specific antibody IgG of every immune group (P〈0. 01). In FCA and Al-adjuvant groups, the antibody IgG reached the peak at the 9th week, and then descend gradually, but in the microsphere group it increased at the 21th week (P〈0. 01); there were significant differences on IL-2 and IFN-γ in microsphere group (P〈0. 05), FCA group (P〈0.01) and Al-adjuvant group (P〈 0.01) at the 6th, 9th week, but in the microsphere group they increased consistently (P〈0.01) from the 12th to 21th week, and in FCA and Al-adjuvant groups they both descended gradually after the 12th week. While the comparison was made between the microsphere group and FCA group as well as the microsphere group and Al-adjuvant group respectively, there were significant differences of serum specific antibody IgG after the 12th week, IL-2 after the 18th week, and IFN-γ after the 15th week in the microsphere group(P〈0. 01). Conclusion There is a longer and higher effect induced by biodegradable microspheres of rSjGST in mice, which provides a new means for further study on a vaccine of Schistosorna japonicurn.
