摘要
目的:观察-4℃纯甘油保存方法对羊膜活性的影响。方法:取健康剖宫产产妇胎盘,剥离羊膜,分成2cm×2cm小块,保存于-4℃纯甘油中,3,7,14,30,60,90d后,分别进行光镜,酶组织化学[乳酸脱氢酶(LDH)、琥珀酸脱氢酶(SDH)]及免疫组织化学[碱性成纤维细胞生长因子(bFGF)]观察,并于新鲜羊膜对比。结果:光镜下,-4℃纯甘油保存60d以内羊膜上皮层结构较完整,可见少数细胞破裂。胞核脱失。90d组上皮层结构破坏明显,多数细胞破裂,核脱失。酶组织化学(LDH、SDH)及免疫组织化学(bFGF)显示:60d内各组于新鲜羊膜比较无显著差异性;各组间两两比较亦无显著性差异(P>0.05)。而90d组于新鲜对照组及各实验组间两两比较均有显著性差异(P<0.05)。结论:羊膜在-4℃纯甘油保存60d内;组织结构;
AIM: To investigate the vitality of amniotic membrane preserved in glycerol at -4℃ .
METHODS: Fresh amniotic membrane (AM) attached to healthy placentas was taken and preserved in glycerol at -4℃. After 3, 7,14, 30, 60, 90d, the epithelial structure (light microscope) and the enzyme activity (SDH and LDH) of AM was analyzed by immunohistochemical method combining with image analysis system to investigate the changes of the expression of basic fibroblast growth factor (bFGF) in AM.
RESULTS: Host of the epithelial cells of AH preserved in glycerol at -4℃ remained intact after 60d. The vitality of AM had a stable level.
CONCLUSION: The vitality of AM preserved at -4℃ glycerol decreased with time. After 60-d preservation, the vitality of AM decreased significantly. Therefore operators can select high-level AM to treat ocular surface diseases.
