摘要
目的:应用SD大鼠下颌骨来源的原代成骨细胞,观察成骨细胞对甲硝唑和替硝唑的转运,探讨人工种植牙给药的影响及可行性。方法:将原代培养的新生鼠成骨细胞(newbornrat’sosteoblast,N)和成年鼠成骨细胞(adultrat’sosteoblast,A)与溶于PBS的药物共同孵育,分别于1,3,5,10min后,弃去细胞外液,收集各时间点细胞悬液,用高效液相色谱法测定细胞内药物量,用考马斯亮蓝法测定细胞蛋白总量。结果:(1)高效液相色谱法可以准确测定2种药物的量。(2)2种成骨细胞均可将甲硝唑和替硝唑转运至细胞内。结论:成骨细胞具有转运硝基咪唑类药物的能力,证实了人工种植牙给药的可行性。
Objective: To observe the transmembrane transport ofmetronidazole and tinidazole by osteoblast from rat' s mandible, and explore the effect and possibility of dental implant administration system. Methods: Co-culture the osteoblasts of newborn(N) or adult(A) rat and agents in PBS at 37℃, remove the extracellular fluid after 1, 3, 5, 10min respectively, collect the suspension, then measure the intra-cellular contents of agents with high performance liquid chromatography(HPLC), and the cell total protein with Coomassie Brilliant Blue. Results: 1, HPLC is an accurate method for the measurement of the intra-cellular contents of the two agents. 2, Adult and newborn rat' s osteoblasts both have the ability of intra-cellular transport of metronidazole and tinidazolet. Conclusions: Osteoblasts from SD rat' s mandible can transport metronidazole and tinidazole into cells, which proves the possibility of the dental implant administration system.
