摘要
从分子水平分析了原产地沙皮犬的遗传背景。以3种类型共25只沙皮犬的血液为材料提取DNA。进行随机扩增多态DNA(RAPD)分析。筛选出的12个重复性、多态性好的随机引物,扩增出96个位点,分子量在350~2000bp,共享带数为17条,多态性82.29%,说明沙皮犬存在丰富的遗传多样性。聚类分析的结果表明:全部骨嘴沙皮犬和大部分肉嘴沙皮犬能分别聚在一块,与其各自的外部形态特征分类基本相符,初步从分子水平为沙皮犬各类型的区分提供了理论依据。
Chinese Shar-pei is one of world famous dogs, which were originated from Dali Town, Naihai district, Foshan City, Guangdeng Province, China. Presently, the quantity of Chinese Shar-pei is very few and the genetic situation of this variety is chaotic. To prevent this precious variety, from extinction, the experimemet in the analysis of the genetic background of this dog variety with RAPD method was firstly conducted. From 25 dogs' blood DNA sangles, 12 random primers were selected from total 71 primers. The electrophoresis results of RAPD showed that 12 primers produced 96 bands with molecular weight ranged 350 - 2000 bp and the shared bands was 17 with polymorphism rate of 82.29%. This showed the rich genetic diversity of the Chinese Shar-pei in its origin place. The clustering .analyses of the RAPD of 25 samples of Chinese Shar-pel showed the bone-mouth and meat-mouth sample could be grouped accordingly,which supplied theoretical basis for the sub-classification of this variety.
