摘要
为了研究干扰素α-2b(IFNα-2b)对HL-60细胞增殖与凋亡的影响,采用瑞氏染色观察HL-60细胞的形态变化,吖啶橙/溴化乙锭双染色观察细胞凋亡,通过CD13测定观察细胞膜抗原表达及成熟分化,MTT实验观察增殖活性。结果显示:加入IFNα-2b48小时后,HL-60细胞凋亡率升高,增殖活性下降,而且随着IFNα-2b浓度的升高,抑制作用更加明显。CD13表达逐渐下降,细胞形态逐渐向成熟转化。结论:IFNα-2b能促进HL-60细胞凋亡、抑制HL-60细胞增殖和促进分化成熟。
To investigate the effects of interferonα-2b on proliferation and apoptosis in HL-60 cells, HL-60 cells were cultured in different concentrations of IFNα-2b. The morphologic changes were observed by Wright' s and acridine orange (AO) and ethidium bromide (EB) staining respectively. Inhibition of proliferation was detected by MTT. Expression of CD13 ^+ was checked by indirect fluoroimmunoassay. The results showed that apoptosis rate of HL-60 ceils assayed by the above-mentioned two methods was (51 ±2)% and (78 ±3)% respectively and OD570 values of proliferation inhibited were 1.8 ±0.1 and 1.0 ±0.1 respectively when the concentrations of the IFN α-2b were 500 and 10 000 U/ml in culture for 48 hours. Morphology and count of CD13^+ cells were changed. CD13^+ cell expression rate was (62 ±2)% and (30±3)% respectively when the concentrations of the IFN α-2b were 500 and 10 000 U/ml in culture for 48 hours. It is concluded that IFN α-2b can enhance the apoptosis of HL-60 cells, inhibit their proliferation, promote their maturation and differentiation.
