摘要
目的探讨苦参碱(Matrine,Mt.)对压力负荷性肥厚心肌TGF-β1的作用。方法应用大鼠腹主动脉部分结扎,建立压力负荷性心肌肥厚模型。实验分四组进行,即:手术组、假手术组、手术苦参碱组、手术卡维地洛(Car.)组。术后5周,观察左心室重量与体重之比(LVW/BW)、用免疫组化法查TGF-β1、以及用RT-PCR法查TGF-β1。结果手术组与假手术组对照,LVW/BW增加;用免疫组化法查TGF-β1,蛋白表达增强;用RT-PCR法查TGF-β1,mRNA表达增强(P<0.05)。手术用药组与手术组比较,LVW/BW减小;用免疫组化法查TGF-β1,蛋白表达减少;用RT-PCR法查TGF-β1,mRNA表达减少(P<0.05)。结论苦参碱具有减少压力负荷性心肌TGF-β1的作用。
Objective To explore the effect of Matrine (Mt.) on myocardial TGF-β1 induced by pressure over-load. Methods Pressure over-loaded myocardial hypertrophy was produced by banding of aorta abdominalis in 67 male Sprague-Dawley rats weighing 200g±15g. The rats were assigned into one of the following groups: sham-operation control, operation control, operation group with treatment (Matrinc 15mg/kg/d and Carvedilol 3.6mg/kg/d). The rats were administered with drugs one day after operation. 5 weeks after treatment, left ventricular weight (LVW), myocardial TGF-β1 was detected by immuno-histochemistry, and mRNA of TGF-β1 was semi-quantified with RT-PCR. Results LVW/BW (body weight), mRNA expression of TGF-β1 (P〈0.05), and TGF-β1 detected by Immuno-histochemistry was obviously increased in operation group compared with sham control group. Mt. and Car. treatment can significantly decrease LVW/ BW (P〈0.05) mRNA expression of TGF-β1 (P〈0.05), and TGF-β1 detected by Immuno-histoehemistry. But there was difference compared with sham group. Conclusions it showed that Matrine can decrease TGF-β1 of hypertrophy cardium induced by pressure over-load.
