摘要
目的:应用分光光度法建立一种简便、准确、灵敏、稳定的重组α-半乳糖苷酶(AGA)活性检测方法。方法:利用紫外-可见分光光度计,根据产物生成量计算所测标本的酶活性。结果:该方法的批内变异系数为2.63%,批间变异系数为4.42%,均〈5%;在所检测该酶质量浓度范围(0.3~2.5)μg/ml)内线性关系良好;每组的平均回收率均在95%~105%之间。结论:与已经建立的测活方法相比较,该方法具有无需酶的标准品,结果可靠、误差小、稳定性好、操作简单,适合实验室及工业化生产应用的优点。
Objectlve:To establish a method that can be used for the precise determination of α-D-galactosidase activity. Method:Products produced by α-D-galactosidase were detected by means of spectrophotometric method, which could be used to evaluate α-D-galactosidase activity. Results:The results showed that the variability within batchs was 2.63% and the variability between batchs was 4.42%. These indicated that the method was reliable to could be used in α-D-galactosidase assay. Conclusion: The method has the merits of high precision, good reproducibility and easy operation, so it can be used for detecting α-D-galactosidase activity exactly.
