摘要
目的研究NF-κB对人眼小梁细胞基质金属蛋白酶-3(matrixmetalloproteinase-3,MMP-3)及组织金属蛋白酶抑制剂-1(tissueinhibitorofmetalloproteinase-1,TIMP-1)mR-NA表达的影响。方法采用原代培养的人眼小梁细胞,通过脂质体转染的方法,将P65表达质粒转染小梁细胞,24h后通过RT-PCR方法检测MMP-3及TIMP-1mRNA的表达。结果P65转染后MMP-3mRNA的表达明显增高,与正常对照组相比差异有显著性(P<0·05),TIMP-1mRNA的表达与正常对照组相比无明显改变(P>0.05)。结论NF-κB的活化可激活MMP-3的表达,提示NF-κB信号通路参与了小梁网外引流通道的调控。
Objective To study the effects of NF-κB on expression of matrix metalloproteinase-3 ( MMP-3 ) and tissue inhibitor of metalloproteinase-1 (TIMP-1 ) mRNA in human trabecular meshwork cells( HTM cells). Methods Lipofectin method was used to transfect P65 plasmid into the primary cultured HTM cells. Twenty-four hours after transfection, the expression of MMP-3 and TIMP-1 mRNA were measured by RT-PCR method. Results The expression of MMP-3 mRNA was dramatically increaser than the control group(P 〈0.05) after transfection,the expression of TIMP-1 mRNA had no significant difference with those in the control group (P 〉 0.05 ). Conclusion The activation of NF-κB can increase the expression of MMP-3 mRNA in HTM cells, which suggests that the signal transduction pathways of NF-κB may participate in the regulation of aqueous humor outflow facility.
