三氯化钆对大鼠肝脏缺血再灌注损伤保护作用的实验研究 被引量:2

Experimental Studies on Protective Effect of Gadolinium Chloride Against Liver Warm Ischemia-reperfusion Injury in Rats

摘要 【目的】探讨枯否细胞抑制剂三氯化钆对大鼠肝脏热缺血再灌注损伤的影响,并分析其可能机制。【方法】将64只成年雄性SD大鼠随机分为3组:三氯化钆预处理+缺血再灌注组(GD组,n=24),生理盐水预处理+缺血再灌注组(NS组,n=24),假手术组(Sham组,n=16)。按Pringle's法建立肝脏95%缺血模型:选择性阻断肝门静脉左支及肝动脉30 min,再灌注120 min。各组经下腔静脉取血测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、肿瘤坏死因子(TNF-α)浓度;各组分别于再灌注120 min后经尾静脉注射印度墨汁,断尾取血测量OD5 minand OD15 min值以计算枯否细胞吞噬系数;取左叶肝组织行病理切片检查。【结果】①缺血30 min及再灌注120 min后GD组ALT、AST、TNF-α浓度低于NS组(P<0.05)。②再灌注120 min后GD组枯否细胞吞噬系数较NS组及Sham组降低(P<0.05)。③GD组再灌注120 min后肝组织形态学改变均较NS组轻。【结论】三氯化钆能减少枯否细胞释放细胞因子(主要是肿瘤坏死因子)从而减轻肝脏热缺血再灌注损伤。 [Objective]To investigate the effect of Kupffer cell inhibitor Gadolinium Chloride(GdCl3) on hepatic warm Ischemia-Reperfusion(IR) injury in rats. [Methods]Sixty four adult male Sprague-Daweley rats were divided randomly into 3 groups: Group A(n= 24) :GdCl3 preconditioning group with IR, Group B( n = 24):normal saline preconditioning group with IR and Group C ( n=16):Sham group. Partial hepatic ischemia model was made by Pringle's method by blocking the left branch of portal vein and hepatic artery selectively for 30min,and reperfused for 120min. Blood sample was obtained through inferior vena cava to measure changes of Tumor Necrosis Factor(TNF-α) and ALT,AST levels. Each group was injected Indian Ink through tail vein at 120min after reperfusion procedure, and the blood sample obtained by cutting off the tails to measure OD 5min and OD 15min for calculation of the phagocytosis index. Liver tissue was obtained from the left lobe to investigate histological changes. [Results]Blood ALT, AST, TNF-α levels in group A were lower than in group B in 30min after ischemia procedure and 120min after reperfusion procedure ( P〈0. 05). Phagocytosis index in group A was lower than Group B and C after 120min of reperfusion procedure( P〈0.05). Intercellular edema ,congestion of hepatic sinusoid and liver cell degeneration in group A was much lighter than in group B. [Conclusion]Gadolinium Chloride can attenuate the liver IR injury,it might result from inhibiting release of cytokines(mainly TNF-α) by Kupffer's cells.
