Establishment and evaluation of oxidative stress models

摘要 目的:总结并评价各类氧化应激模型的建立及优缺点,探讨建立最佳的氧化应激模型,以进一步深入研究氧化损伤的机制以及开发新的抗氧化应激药物。资料来源:应用计算机检索Pubmed1994-01/2005-12关于氧化应激的文章。检索词“oxidativestress”并限定文章的语种类为English。同时利用计算机检索中国期刊全文数据库1994-01/2005-12的相关文章,限定文章语言种类为中文,检索词“氧化损伤”。资料选择:对资料进行初审,纳入标准:①关于氧化应激模型的成因及其建立的方法。②氧化应激模型的应用研究。排除标准:排除重复性研究。资料提炼:共收集到符合上述要求的文献40篇,排除23篇重复性研究,纳入17篇。资料综合:实验表明,氧化应激在一些疾病的发生发展中起着非常重要的作用,建立氧化应激模型有着非常重要的实际意义。现有氧化应激模型主要有动物模型和细胞培养模型两种形式(也可称为在体模型和离体模型),其中前者又可分为特异性和非特异性两类模型。动物模型中以D-半乳糖模型应用最广泛,而细胞模型中以过氧化氢氧化损伤模型为著。结论:在活体和离体培养细胞上建立多种氧化应激模型,各种模型均有其优缺点,为建立稳定、确切的氧化应激模型,进一步研究开发新的抗氧化应激药物,应将多种模型有效结合。 OBJECTIVE: To summarize and evaluate all sorts of oxidative stress models and their advantages and disadvantages, and approach the optimal oxidative stress model, so as to further study the mechanisms of oxidative stress and develop new antioxidants. DATA SOURCES: A computer-based online search of Pubmed was undertaken for articles about oxidative stress published in English between January 1994 and December 2005 with the key words of "oxidative stress". Meanwhile, we searched the China Journal Full-text Database for related articles published in Chinese between January 1994 and December 2005 with the same key words in Chinese. STUDY SELECTION: The literatures were selected primarily. Inclusive criteria: (1)Causes and methods for establishment. (2) Application of oxidative stress models. Exclusive criteria: repetitive studies. DATA EXTRACTION: There were 40 literatures in accordance with the inclusive criteria, 17 ones were selected, and 23 repetitive ones were excluded. DATA SYNTHESIS: According to experiments, oxidative stress plays a very important role in the pathogenesis and etiologies of some diseases. So it is very important to establish the models of oxidative stress. Current oxidative stress models mainly include animal models and cell models (also named ex vivo and in vivo models), and the former comprises specific and non-specific models. D-galactose model is extensively applied among animal models. And H202-induced oxidative damage model is popular among ex vivo models. CONCLUSION: Oxidative stress models can be made in vivo and vitro, both of them have benefits and limitations. To establish stable and certain oxidative stress models and develop new antioxidants, all sorts of models should be integrated together.
