速降糖对链脲佐菌素糖尿病大鼠心肌组织AT_1的影响

摘要 目的:观察速降糖对链脲佐菌素(STZ)糖尿病大鼠血糖及心重指数、心肌组织血管紧张素Ⅱ受体1(AT1)表达的影响,探讨速降糖对糖尿病(DM)大鼠病变早期心肌保护作用的机制。方法:通过腹腔注射STZ建立DM大鼠模型,正常组、模型组给生理盐水,大小剂量组给225%、75%速降糖合剂,西药Ⅰ组给二甲双胍(1.5%),西药Ⅱ组给二甲双胍(1.5%)与氯沙坦(0.05%)混悬液灌胃,每日1次,连续8周。结果:与正常组比较,模型组体重降低(P<0.01)、血糖升高(P<0.01)、心重指数增加(P<0.01)、AT1的表达显著增加。治疗组体重高于模型组、血糖低于模型组(P<0.05或P<0.01)、心重指数下降(P<0.01)、AT1表达降低,以大剂量组和西药Ⅱ组最为显著。结论:速降糖对DM大鼠病变早期心肌有良好保护作用,并且与剂量呈正相关。 Objective:To observe the effect of Gliquidone on blood glucose, cardiac weight index and the expression of ATlin cardiac muscle tissue of DM rats induced by STZ and to discuss the mechanism of Gliquidone's protection on cardiac muscle in early stage of DM rats. Method: Modeling DM rats are with STZ in intrperitoneal injection. Normal group and model group were seeded in Sodium Chloride. Large and small dose groups were seeded in 225 % ,75% Gliquidone mixture separately. Western medicine group Ⅰ were seeded in Metformin (1. 50%) while western group Ⅱ were seeded in suspl. Met romin (1.5%) and Losartan (0.05%) by intragastric administration. It lasted 8 weeks and once a day. Result: Compared with the normal group, the model group's weight decreases(P〈 0.01) ,blood glucose advances (P〈0.01), cardiac weight index rises (P〈0.01) and the expression of AT1 increases significantly. The weight of the therapeutic group is higher than that of the model group while the blood glucose of the therapeutic group is lower than that of the model group (P〈0.05 or P〈0. 01). The therapeutic groups" cardiac weight index decreases (P〈0.01) and the expression of AT 1 depresses. The change of large dose group and western medicine group Ⅱ is the most obvious. Conclusion.. Gliquidone has good protection on cardiac muscle in early stage of DM rats, which is positively correlated with the dose.
