摘要 目的通过对马鹿卵母细胞的超微结构进行研究,旨在为马鹿繁殖育种的研究提供基础理论依据。方法用FSH对母马鹿进行超数排卵,采用抽吸法和切割法采集卵母细胞,利用透射电镜观察其超微结构。结果在光镜下,马鹿卵母细胞的平均直径为172.958±17.298μm,卵丘细胞厚度为67.750±33.107μm,透明带厚度为13.782±2.516μm。电镜下,卵母细胞外的卵丘细胞紧紧包裹卵母细胞,且与卵母细胞之间有大量细长的微绒毛相联系;卵丘细胞核呈卵圆形,其长径为4.250±0.042μm,短径为2.750±0.071μm;透明带均匀,与卵母细胞结合紧密;在皮质区集中分布大量的多嵴型线粒体、脂滴和空泡,线粒体长径为0.600±0.106μm,短径为0.490±0.117μm;细胞核中颗粒状的核质分布非常均匀。 Objective To study the ultrastructural changes of wapiti oocytes and provide theoretical basis for wapiti breeding and reproduction. Methods The oocytes for ultrastructures observation under the transmission electron microscope were from the deer dealt with FSH for superovulation. Collecting methods were aspiration and deeply-cut. Results Under the fight microscope, we had figured out that the mean diameter of wapiti oocyte was 172.958 ± 17.298 μ m, and the thickness of cumulus cell and zona pellucida were 67.750 ± 33.107 μ m and 13.782 ± 2.516 μ m respectively. Under the transmission electron microscope,we had observed that the state of oocytes and the cumulus cells around them was tightness, and there were plenty of long and thin microvilli playing the role of connection m the enwrapping. The shape of cumulus ceils nucleus was ovate, and the longer and shorter diameters of the cumulus ceils were 4.250 -± 0.042 μm and 2.750 _±0.071 μm respectively. The zona pellucida was well proportioned and tighdy linking with oo .cyte. There were great deals of multi-crista mitochondria,lipid droplet and vacuolus. The mitochondria longer and shorter diameters were 0.600 _± 0.106 μ m and 0.490 ± 0.117 μ m. Granular nucleoplasma distributed very well in the ceil nucleus.
