摘要
本文合成了二个模拟阿维菌素(AvermectinB_1)特征结构的半抗原,4″-O-(单)琥珀酰阿维菌素和5-O-(单)琥珀酰阿维菌素。将半抗原与载体蛋白(BSA、OVA和KLH)连接,制成人工抗原。用4″-O-(单)琥珀酰阿维菌素-BSA免疫家兔,制备针对阿维菌素的多克隆抗体。以间接ELISA方法对抗体进行了鉴定。
The polyclonal anti-Avermectin B_1(AVM)antibodies were developed. 4″-0-succinoyl AVM(hapten)and 5-O-succinoly AVM(hapten)were synthetized successfully. The haptens were coupled to BSA, OVA and KLH by NHS ester procedure. The antibodies were obtained successfully by immunizing New Zealand rabbits with 4″-0-succinoyl AVM-BSA.The indirect competitive ELISA for AVM was established. The antibodies had high specificity and avidity for AVM(I50=5.6ng/ml),and can also recognized Ivermectin(I50=10.0ng/ml).
