摘要
目的:他汀类药物的降脂作用已为人们熟知,近几年,其降脂外作用备受关注。总结他汀类药物的降脂外作用,分析并探讨其非降脂作用机制,为他汀类药物的临床应用提供了更广阔的前景。资料来源:应用计算机检索PUBMED1998-01/2004-12关于他汀类药物非降脂作用的文章。检索词“statin,non-lipideffects”并限定文章语种为English,同时利用计算机检索中国期刊全文数据库2000-01/2004-12相关文章,限定语种为中文,检索词为“他汀类药物,非降脂作用”。资料选择:对资料进行初审,纳入标准:①关于他汀类药物非降制作用的机制及其应用。②对其中某一作用的实验研究。排除标准:排除重复性研究及重复性综述。资料提炼:共收集到符合上述要求的文章58篇,排除36篇,22篇符合纳入标准。其中24篇关于他汀类药物非降制作用的实验研究,3篇关于他汀类药物非降制作用机制及应用的分析。资料综合:他汀类药物非降制作用主要与以下几方面有关:心理健康、骨质疏松、脑卒中、神经炎症性疾病、外周动脉疾病、冠心病、心律失常、心力衰竭、老年斑。这些非降脂作用与其分子生物学机制与药理学机制密切相关。结论:他汀类药物除降脂作用外,还有很多非降脂作用,其非降脂作用应用于临床许多领域。
OBJECTIVE: Statins have been known as a lipid-lowering therapy, and in recent years, people concern more about the non-lipid effects of stafins. On the basis of national and international materials, the paper aims to summarize the non-lipid effects of statins and analyze the mechanism in order to extend their clinical application.
DATA SOURCES: The relevant articles to non-lipid effects of statins between January 2000 and December 2004 were searched in China Journal Full-text Database (CJFD) by imputing the key words of "statins, non-lipid effects" in Chinese into computer; Similarly, the relevant English articles between January 1998 and December 2004 were searched in PUBMED with the key words of "statins, non-lipid effects".
STUDY SELECTION: The literatures about non-lipid effects of statins were selected primarily. Inclusion criteria: (1) The mechanism and application of statins' non-lipid effects.(2)Experimental study of a certain effect. Exclusion criteria: Repetitive research or review.
DATA EXTRACTION: There were 58 literatures in accordance with the criteria, and 36 of them were excluded due to the repetitive researches. Among the 22 selected literatures, 24 were experimental studies of statins' non-lipid effects and 3 were analysis about the mechanism and application of this effect.
DATA SYNTHESIS: The non-lipid effects of statins are mainly related with: psychological well-being, osteoperosis, neuro-inflammatory disorders, ischemic stroke, peripheral arterial diseases, coronary heart diseases, arrhythmias, heart failures and age-related maculopathy, as well as molecular biological mechanism and pharmacological mechanism.
CONCLUSION: Despite of the cholesterol-lowing therapy, statins are available in many clinical realms for non-lipid effects.
