高效液相色谱法测定六维胶丸中维生素B_1、维生素B_2、烟酰胺含量 被引量:13

摘要 目的:建立高效液相色谱法测定六维胶丸中维生素B1、维生素B2、烟酰胺的含量。方法:采用Agilent 1100高效液相色谱仪,C18。柱(250mm×4.6mm,5μm)为色谱柱;流动相为离子对溶液(含1.5mmol/L戊烷磺酸钠和3.4mmol/L庚烷磺酸钠的0.24%三乙胺溶液,用冰醋酸调节pH值至3.0)-甲醇(80:20),检测波长254nm,流速l,0mL/min。以外标法检测定量。结果:维生素B,的线性范围是4.84~24.2μg/mL,r=0.999674,平均回收率为1.03.3%,RSD=2.0%;维生素B:的线性范围是6.28~31.4μg/mL,r=0.999915,平均回收率为99.5%,RSD=0.7%;烟酰胺的线性范围是35.3~155.32μ/mL,r=0.999917,平均回收率为97.8%,RSD=0.9%。结论:本方法分离度佳、灵敏度高、重现性好、结果可靠。 Objective :To establish a high-pressure liquid chromatography (HPLC) method for determination of the concentrations of Vitamin B1, vitamin B2, nicotinamide in Liuwei Jiaowan Hexavitamin Soft Capsules. Method: HPLC system ( Agilent 1100 ) was used. The analytical column was packed with C18 (250mm×4.6 mm ID, 5μm). The mobile phase, consisted of ion pair solution (1.5 mmol/L sodium 1 -pentanesulfonate, 3.4 mmol/L sodium 1 - heptanesulfonate and 0.24% triethylamine, pH = 3.0 adjusted by acetic acid)-methyl alcohol ( 80:20, v/v ). The detection was operated at 254 nm. Flow rate was set at 1.0 mL/min. The concentrations were quantified by external standard method. Results: The concentration of Vitamin B1 was linear in the range of 4.84-24.2μg/mL (r=0.999 674 ). The average recovery rate was 103.3% ( RSD=2.0% ). The concentration of Vitamin B2 was linear in the range of 6.28-31.4μg/mL ( r = 0. 999 915 ). The average recovery rate was 99.5% ( RSD = 0.7% ). The concentration of nicotinamide was linear in the range of 35.3-155.32 μg/mL (r=0.999917 ). The average recovery rate was 97.8%(RSD=0.9%). Conclusion : Vitamin B1 , Vitamin B2 and nicotinamide are completely separated under this chromatographic condition. The analytical method shows high sensitivity and good reproducibility. It is proved to be. reliable.
