摘要
环氧合酶(COX)是花生四烯酸代谢途径的重要限速酶,其代谢产物在血小板聚集、炎症反应等心血管病理生理过程中发挥重要作用。近年来有关COX-2抑制剂心血管安全性的争论使得COX在心血管疾病中的作用越来越受重视。现就COX及其同工酶、COX在动脉粥样硬化、血栓形成、心肌缺血、心力衰竭等心血管疾病发生中的作用以及COX-2抑制剂心血管安全性等方面的进展作一综述。
Cyclooxygenase (COX) is one of the main rate-limiting enzymes in the conversion of arachidonic acid into prostaglandins. COX plays an important role in a variety of cardiovascular pathophysiological processes, such as platelet aggregation and inflammatory re- sponse. Recently, the controversy about the cardiovascular safety of selective COX-2 inhibitors has led to increased study on the action of COX in the pathogenesis of cardiovascular diseases. This article summarizes recent developments research into COX isozymes, actions of COX in atherosclerosis, thrombosis, myocardial ischemia, heart failure and the cardiovascular safety of selective COX-2 inhibitors.
