摘要
差异蛋白质组学是蛋白质组学的研究策略之一,主要目标是研究有意义的因素引起的蛋白质组成的变化以发现有差异的蛋白质。介绍了差异蛋白质组学应用于植物研究中的基本方法和新兴技术。植物样品制备方法主要为TCA/丙酮法和植物组织分级提取法。2-DE仍是目前主要的分离方法,多维液相色谱等新型分离模式可以弥补2-DE的不足,新型质谱和蛋白质芯片技术的应用可大大提高差异蛋白质的鉴定速度。差异蛋白质组学目前在植物生理、植物组织器官和亚细胞、植物突变体、植物遗传多样性等多方面研究中都有应用。
The differential preteomic is one of preteomic's rescareh strategies, which mainly aims to discover the difference protein based on the rescareh on the protein composition change caused by significance factor. In the article the essential methcod and new technology used to research differential proteomics in the plant were introduced. The preparation method of plant sample was mainly TCA/acetone strategy and plant organization graduation extraction pmocess.2-DE was still the main separation method at present. The new separation pattem such as multi-dimenional liquid chromatography may make up 2-DE's insufficiency. The new technology application of mass spectrometry and the protein chips may enhance aooraisal speed of the difference protein greatly.
