摘要
目的探讨血管内皮生长因子(VEGF)对脊髓损伤细胞凋亡的影响及保护机制。方法采用Allen的Weightdropping法挫伤大鼠T10节段脊髓,于术后2、4、8h,1、3、7d经蛛网膜下隙导管各注入VEGF溶液(5μl/100g),并与生理盐水组和正常组作对照。采用原位末端标记法标记脱氧核糖核酸片段,检测脊髓损伤前后发生凋亡的脊髓细胞。结果正常对照组大鼠脊髓中未见凋亡细胞。生理盐水组于伤后4h始出现凋亡细胞。VEGF组与生理盐水组相比较,凋亡细胞明显减少(P<0.01)。结论VEGF可抑制脊髓损伤后细胞的凋亡,从而保护损伤的脊髓组织。
Objective: To explore the effect of vascular endothelial growth factor (VEGF) on cell apoptosis after spinal cord injury (SCI) and the proteetive meehanism of VEGF in injured spinal eord. Methods:Fifty-throe Wistar rats were randomly divided into 3 groups :eontrol group ( receiving lamineetomy; n = 5 ), saline group ( subaraehnoid injection of saline ; n = 24 ) , and VEGF group ( subaraehnoid injection of VEFG of 5 μl/100 g; n =24). The SCI rat model was established by using Allen' s weight dropping method, and the thoracic vertebra (T10) was injured. The apoptotie eells were detected by TUNEL staining 2, 4, and 8 hours and 1, 3, and 7 days after SCI. Results:No apoptotie eell was found in eontrol group. The apoptotie eells were found 4 hours after SCI in saline group. Compared with saline group, the number of apoptotie eells was signifieandy less in VEGF group at eaeh time point ( P 〈 0.01 ). Conelusion:VEGF may proteet injured spinal eord by suppressing the eell apoptosis after SCI.
