用于定量药物脑电图疼痛程度监测的肌松兔模型 被引量:16

摘要 目的:为定量药物脑电图监测疼痛程度建立肌松兔模型。方法:实验于2005-07/08在徐州医学院麻醉学基础教学实验室完成。选择健康壮年中国家兔18只,雌雄不拘,随机数字表法分为擦拭组和切皮组,每组9只。30g/L戊巴比妥钠30mg/kg静脉麻醉和维库溴铵0.3mg/kg建立肌松兔模型肌肉松弛后,进行机械通气,频率30次/min,潮气量10mL/kg。擦拭组给予非伤害性刺激,干棉球擦拭腹正中皮肤10cm;切皮组给予伤害性刺激,做腹正中腹膜外切口10cm。观察刺激前后兔定量药物脑电图各频段功率百分比的变化。脑电图的处理是根据其频率分为δ、θ、α1、α2、α3、β6个频段,记录同一脑区不同频段所占功率百分比。频段划分:δ(0.8~3.8Hz)、θ(4.0~7.8Hz)、α1(8.0~8.8Hz)、α2(9.0~10.8Hz)、α3(11.0~13.0Hz)、β(13.0~30.0Hz)。采集刺激前30s及刺激后60s的脑电信号,每次采样5s。结果:18只家兔均进入结果分析,无脱失值。①与刺激前相比,擦拭组各频段功率百分比无明显改变(P>0.05);切皮组除θ频段外,各频段在不同脑区均有不同程度的改变(P<0.05)。②各组变异系数低于60%。擦拭组变异系数在20%~50%之间,切皮组变异系数在20%~60%之间。结论:肌松兔模型稳定、可靠,可用于镇痛程度的定量药物脑电图监测研究。 AIM:To set up a kind of rabbit model with muscle relaxant for usage of quantitative pharmaco-electroenciphalogram (QPEEG) in monitoring pain. METHODS: The experiment was performed at the Basic Laboratory of Anesthesiology, Xuzhou Medical College from July to August 2005. Totally 18 healthy adult rabbits (Chinese rabbit), of either sex, were randomly divided into 2 groups (9 rabbits in each group): mopping group and cutting group. Different stimulation were given to the rabbits which had been injected 30 g/L pentobarbital sodium 30 mg/kg and vecuronium bromide 0.3 mg/kg by vein, and performed mechanical ventilation with the frequency of 30 R/min, tidal volume (VT) of 10 mL/kg. The mopping group received non-harmful stimulation (central skin 10 cm of abdomen was cleaned with dry cotton ball) and the cutting group received harmful stimulation (external incision 10 cm of central peritoneum was made). The percentage of power of each frequency band of QPEEG sample was obtained before and after the stimulation. The EEG was divided into six bands including δ (0.8-3.8 Hz), 0 (4.0-7.8 Hz), α1 (8.0-8.8 Hz), α2 (9.0-10.8 Hz), α3 (11.0-13.0 Hz) and 13(13.0-30.0 Hz). The QPEEG 30 s before and 60 s after stimulation were recorded. The sampling action time was 5 s on each time point. RESULTS: Totally 18 rabbits were involved in the result analysis, drop-out.①The percentage of every band power showed no significant changes in mopping group (P 〉 0.05); All figures, expect for θ band changed significantly to different degree in cutting group in different areas of brain (P 〈 0.05).②The coefficient of variation of the figures were lower than 60%.The coefficient of variation of mopping group was between 20%- 50%, and cutting group was between 20%-60%. CONCLUSION: The model is stable and reliable, which can be used in the study of monitoring pain with QPEEG.
