摘要
目的观察血府逐瘀口服液对大鼠溃疡性结肠炎(ulcerative colitis,UC)模型的治疗作用,探讨血栓前状态(prethrombosis state,PTS)在UC发病中的作用。方法用2,4,6-三硝基苯磺酸(2,4,6-trinitrobenzenesulfonic acid,TNBS)制备大鼠UC模型,将动物模型随机分为模型组,模型+生理盐水组,模型+血府逐瘀口服液组,另设空白对照组,共4组。观察各组大鼠肠道大体形态和组织学改变;检测血小板黏附功能(platelet adhesivability,PAdT)和血小板聚集功能(platelet aggregability,PAgT);酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)测定血清中的血栓烷B2(thromboxane B2,TXB2)、血管性假血友病因子相关抗原(von Willebrand Factor antigen,vWF∶Ag)和细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)。结果与模型+生理盐水组比较,模型+血府逐瘀口服液组大体形态和组织学损伤指数明显下降(6.02±0.74 vs 0.93±0.36,P<0.01;5.68±0.60 vs 1.13±0.32,P<0.01);模型+血府逐瘀口服液组的PAdT和PAgT较模型+生理盐水组降低(P<0.05);模型+血府逐瘀口服液组的TXB2、vWF∶AgI、CAM-1均较模型+生理盐水组明显降低(P<0.01)。结论TNBS所诱导大鼠UC模型与PTS有一定关系。血府逐瘀口服液通过抗血小板聚集、抗炎症介质而发挥治疗UC作用。
Objective To investigate the effects of xuefuzhuyu, a Chinese oral liquid preparation（XFZY）on rat model of ulcerative colitis（ UC） and to study the role of prethrombosis state（ PTS） on the pathogenesis of UC. Methods The UC model rats induced by TNBS were divided randomly into model group（M）, saline treated group（NS）, XFZY treated group（XFZY）, and blank control group（N）. The changes of morphology and histology of rat intestinal tracts were observed, then platelet adhesivability（PAdT）and platelet aggregability（PAgT）were detected by equipment and the concentration levels of TXB2,vWF：Ag and ICAM-1 in serum were detected by enzyme-linked immuno-sorbent assay（ELISA）. Results In comparison with group NS, the morphologic and histological injury index of XFZY group decreased significandy （6.02±0.74 vs. 0.93±0.36 ,P〈0.01;5.68±0.60 vs 1.13±0.32,P〈0.01）; PAdT and PAgT of XFZY group decreased significantly than those in NS group （P〈0.05）; TXB2, vWF：Ag and ICAM-1 of XFZY group decreased more significantly than those in NS group（ P〈0.01 ）. Conclusion PTS may play a role in the pathogenesis of UC rat model induced by TNBS. XFZY exerts its therapeutic effects through anti-agglomeration of platelets and inhibition of release of inflammatory chemokines and cytokines.
