摘要
目的:本研究通过玉屏风散治疗对卵白蛋白(OVA)致敏和激发的过敏性鼻炎小鼠模型,了解其对Th1和Th2及其平衡的影响。方法:8周龄Balb/c小鼠经OVA全身和鼻部局部致敏激发制作过敏性鼻炎小鼠动物模型,早期干预组和治疗组分别给予玉屏风散提取物2,500 mg/kg,7天后采用流式细胞术分别检测脾脏Th1和Th2细胞以及鼻部病理改变。结果:动物模型组Th2细胞表达显著增高(9.86±1.40);玉屏风散治疗组Th2表达显著下降(3.41±0.72,P<0.01);中药干预组也可保护OVA诱导的Th2细胞早期表达增高(6.27±1.21,P<0.05)。本研究表明玉屏风散可抑制Th2介导的上气道非特异性炎症反应(F=30.53,P<0.0001)。结论:本研究证明玉屏风散对Th2介导的上气道非特异性炎症反应有治疗作用,该方剂能提高Th1/Th2比值和抑制Th2细胞的过度表达。
AIM: To study the effect of Yupingfeng Powder(YPFP) on ratg allergic rhinitis induced by stimulating ovalbumin, and reveal the influence on Thl ,Th2 and Th1/Th2 proportion. METHODS: 8-week-old BALB/c mice were sensitized by means of intranasal and systemic intraperitoneal injection application of OVA, 6 subjects were administered including 2,500 mg/kg extracts of YPFP for 7 days in early stage( interference group) ; 6 mice were administered YPFP in after challenge (therapy group) ; the placebo group were given saline. After 7 days, Thl and Th2 in splenocyte were detected by flow cytometry (FCM) and nasobuccal mucosa pathology were observed. RESULTS: When compared with Th2 of animal model group (9.86 ± 1.40) , there was a significant decrease expressing Th2 after PFP treatment ( 3.41 ±0.72, P 〈 0.01 ), but interference group was also significant protection against the OVA-induced Th2 early response ( 6.27±1.21, P 〈 0.05 ), This study could demonstrate that YPFP inhibited Th2-induced upper airway inflammation ( F = 30.53, P 〈 0. 0001 ). CONCLUSION: The present study provides a first insight into YPFP might therapeutically affect Th2 dominant airway inflammation,clearly indicating that YPFP can elevate ratio of Th1/Th2 and suppress expression of Th2.
