摘要
目的建立测定花芪通脉颗粒中灯盏乙素含量的方法。方法采用反相高效液相色谱法RP—HPLC,ZORBAX-C18(4.6mm×150mm,5μm)色谱柱,甲醇-0.01mol·L^-1 磷酸缓冲液PH为5.2(35:65)为流动相,检测波长335nm,流速0.8ml·min^-1,枉温25℃。结果灯盏乙素在0.74—3.70μg范围内与峰面积呈良好线性关系,r=0.9996,灯盏乙素平均加样回收率为97.4%,RSD为0.98%(n=5)。结论:该方法可靠性高,准确性和重现性好,适用于花芪通脉颗粒中灯盏乙素的含量测定。
Objective To establish a simple and rapid reversed -phase high performance liquid chromatography method for the baseline separation, identification and assay of scutellarin in Huaqi Tongmai Granula. Methods The analytical column was a ZOD- BAX-CIs (4.6 mm × 150 mm,5 μm)filled with a 5 mm stationary phase. The mobile phase Consistent of methanol - (0.01 mol - L^-1 )phosphate solution PH5.2(35:65). The flow - rate was 0.8 ml·L^-1. The injection volume was 20 μl. The Detector wavelength was set at 335 nm, and the column temperature was 25℃. Results The calibration curve was linear over the range of 0.74 - 3.7 μg( r = 0. 999 6). The average recovery of the method was 97.4%, RSD was 0.98% (n = 5 ). Conclusion The proposed method would be used in the quantitative analysis of scutellarin in Huaqi Tongmai Granula and the method is convenient,economical and accurate with good reproducibility.
