摘要
建立反相高效液相色谱法同时测定了杜仲叶中2种黄酮类成分——芦丁和槲皮素的含量。色谱柱为Nova-Pak C18(3.9 mm×300 mm,4μm),流动相为甲醇-0.5%磷酸溶液(505∶0),流速0.6 ml/min,检测波长为356 nm。芦丁和槲皮素的线性范围分别为0.246~4.92μg(r=0.999 93)和0.0846~1.692μg(r=0.999 89),平均回收率(n=5)分别99.8%和102.1%。方法准确、快速、重现性好,适用于同时测定杜仲叶中黄酮类成分的含量。
A RP- HPLC method for the separation and simultaneously determination of two flavonoids,rutin and quercetin in Eucommia ulmoides leaves was established.The separation was performed on a Nova-Pak C18 column (3.9 mm ×300 mm,4μm). Methanol -0.5 % H3PO4 (50:50) was used as the mobile. The flow rate was 0.6 ml/min. The detection wavelength was at 356 nm. C, ood linearities of rutin and quercetin were obtained over the ranges of 0.246-4.92 μg(r= 0.999 93 )and 0.084 6-1.692 μg( r=0.999 89 ). The average recoveries(n=5 )were 99.8 % and 102.1%, respectively. This method is accurate, rapid and reproducible, can be used to determine flavonoids in the material of Eucommia ulmoides.
