摘要
目的探讨在制动法复制骨性关节炎动物模型中,过屈位固定法的可靠性和优越性。方法将6 月龄的健康普通级新西兰大耳兔36只分成2组,分别将左后肢膝关节过屈位(A 组)和过伸位(B 组)固定,于固定后第7周、13周分别宰杀取材,比较两组在大体解剖学观察、软骨组织病理学评分、软骨细胞凋亡指数等指标的变化。结果 A、B 两组软骨退变符合 OA 病理特征,过屈位组在大体解剖学观察、软骨组织病理学评分,软骨细胞凋亡指数等指标上略轻于过伸位组,但两组比较无显著性差异(P>0.05)。结论在相同的制动时间里,过伸位和过屈位都能能成功的复制出 OA 动物模型,但过屈位法具有简便、固定牢靠、耗材少、家兔适用性好、成功率高的优点。
Objective To evaluate the reliability and superiority of immobilization method in bend excessive position among immobilization methods to reduce osteoarthritis animal model. Methods 36 six-month-old New Zealand rabbits were randomly divided into two groups. The left posterior knee joints in group A were fixed in bend excessive position, and group B in extend excessive position. Half of each group were decapitated in the 7th weeks and 13th weeks respectively. The gross morphological condition was observed and histopathology score was graded by C. Colombo criteria. And apoptosis ofchondrocytes was detected by immunohistochemical method. Results The findings of histological study showed that degenerative changes in both group A and group B ac- corded with character ofosteoarthritis, and there was not significant difference in indexes such as C. Colombo score and the chondrocytes apoptosis index (P〉0.05) . Conclusion Both methods can successfully induce the animal model of osteoarthritis in the same immobilization time, but the bend excessive position immobilization method has its superiority, for example: simpleness, sturdy fixation, fewer waste, better applicability and higher succeed rate.
