摘要
目的 测定金菊花中绿原酸的含量。方法 采用waters 515高效液相色谱仪,色谱栓(4μm,4.6mm×250mm),流动相为0.1mol/L磷酸二氢钠的缓冲溶液(pH2.65)-甲醇(76:24);流速1.0ml/min;柱温为室温;检测系统waters2487紫外检测器;检测波长为328nm。结果 绿原酸浓度线性范围在0.095-0.475μg内与峰面积呈良好的线性关系,相关系数为0.9999。结论 该法操作简便。准确,重复性好,可作为全菊花的定量分析方法。
Objective To establish the method for determining the content of chlorogenic acid in Dendranthema morifolium with HPLC. Methods The column with acetonitfile-1%acetic acid solution(76: 24)as the mobile phase was used. The flow rate was 1.0ml/min and the detective wavelength was 328 nm. Results The calibration curve was linear at a range of 0.095 - 0.475 μg for baicalin(r =0.999 9). Conclusion The method is simple and accurate with a good reproducibility,and can be used as quantitative analysis method for this preparation.
