摘要
目的初步探讨蚯蚓与日本血吸虫免疫交叉反应的分子机制,为研究和解决临床常见寄生蠕虫病相互间免疫交叉反应现象提供理论和实验依据.方法采用噬菌体展示技术(PDT)、饱和硫酸铵溶液梯度盐析法、SDS-PAGE和Western blot,对经12肽库筛选所获蚯蚓与日本血吸虫共同模拟表位以及蚯蚓的盐析沉淀蛋白、蚓激酶和热刺激分泌物的分子成分及免疫活性进行研究.结果获得了3个蚯蚓与日本血吸虫的共同模拟表位,分别包含NSIL、LAET和HT-HI氨基酸序列.蚯蚓与日本血吸虫的共同模拟表位以及蚯蚓的盐析沉淀蛋白、蚓激酶及热刺激分泌物均存在分子量为28.3~97.8 kDa的蛋白带,其中能被日本血吸虫病患者血清识别的反应带对应分子量在61.2~95.1 kDa.结论蚯蚓与日本血吸虫间存在分子量为61.2~95.1 kDa的共同分子成分,该成分中可能部分属于酶类,氨基酸序列可能包含NSIL、LAET和HT-HI片段,该类物质是蚯蚓与日本血吸虫免疫交叉反应的重要分子基础,由此推测,这也许是蠕虫病相互间免疫交叉反应的重要分子基础.
Objective To explore the molecular mechanism of immune cross-reaction between Lumbricus terrestris and Schistosoma japonicum, and provide theoretical and experimental basis for the research on immune cross-reactive phenomena among the clinical parasitic helminthiasis. Methods Phage display technique (PDT), the salting out method, SDS-PAGE and Western blot were used to study the molecular components and immunocompetence of the common epitopes between L. terrestris and S. japonicurn by PDT, depositions by salting out method, lumbrukinase and secretion with hyperthermia from L. terrestris. Results Three common epitopes between L. terrestris and S. japonicum were obtained, which had NSIL, LAET and HT-HI sequences respectively. The common epitopes, depositions, lumbrukinase and secretion all had the protein bands with molecular weight of 28.3- 97.8 kDa, in which the bands with molecular weight of 61.2- 95.1 kDa were recognized by the sera from schistosomiasis patients. Conclusions The common molecular components with molecular weight of 61.2-95.1 kDa between L. terrestris and S. japonicum may be enzyme and have NSIL, LAET and HT-HI sequences. These components play an important role in the immune cross-reaction between L. rerrestris and S. japonicum, and perhaps are the important molecular basis of immune cross-reaction among the parasitic helminthiasis.
