Establishment of gene transformation acceptor system of cullus in Lilium

摘要 以东方百合“索邦”(L ilium tenu if olium orien ta l‘Sorbonne’)的鳞片、再生形成的小鳞片和叶柄为外植体,进行了愈伤组织诱导、不定芽分化、潮霉素抗性筛选及生根研究。结果表明,3种外植体均可在培养基M S+0.5 m g/L NAA+0.4 m g/L 6-BA+90 g/L Sucrose+4.0 m g/L VB1中形成大量的愈伤组织,愈伤组织分化不定芽的最适培养基为M S+0.5 m g/L 6-BA,筛选的潮霉素质量浓度为20 m g/L,生根培养基为1/2M S+0.2或0.4 m g/L IBA+0.1%活性炭。 Experiments of the callus induction,regeneration of adventitious buds ,the sensibility ol callus to hygromycin and rooting were conducted with the scale leaf,the regenerated scale leaf and petioles as explants. The results showed abundant calluses could be induced from the three explants by the same media. The combination of MS+0.5 mg/L NAA+0.4 mg/L 6-BA+90 g/L sucrose+4.0 mg/L VB; was the optimum medium for the induction of calluses ;the medium of MS+0.5 mg/L 6-BA was the best medium for regeneration adventitious buds from callus,and the medium of 1/2 MS+0.4 mg/L 1BA+0. 1 % active carbon was the best for rooting.
