摘要
目的探讨天麻钩藤饮对肾血管性高血压大鼠(RHR)心肌纤维化的效应。方法采用经典的肾动脉钳夹方法建立RHR模型,观测大鼠胶原浓度,Ⅰ、Ⅲ型胶原含量,心脏重量,左心室重量。结果1.天麻钩藤饮和巯甲丙脯酸均能明显降低肾血管性高血压大鼠左心室重量指数(LVI)水平和胶原的含量(P<0.01)。2.积分法显示天麻钩藤饮组和巯甲丙脯酸组的Ⅰ型胶原含量明显较模型组减少(P<0.05,P<0.01)。模型组和巯甲丙脯酸组的Ⅲ型胶原含量明显较假手术组增高(P<0.01),天麻钩藤饮组的Ⅲ型胶原含量与假手术组比较无明显差异。结论1.天麻钩藤饮与巯甲丙脯酸均能通过降低肾血管性高血压大鼠LVI水平和胶原的含量来干预心肌纤维化。2.巯甲丙脯酸通过降低Ⅰ型胶原的含量,达到降低胶原浓度的作用来干预心肌纤维化;而天麻钩藤饮可同时降低Ⅰ、Ⅲ型胶原的含量,达到降低胶原浓度的作用,从而干预心肌纤维化。
Objective To investigate the effect of Tianma Gouteng Decoction on myocardial fibrosis in renovascular hypertensive rat (RHR). Methods RHR models were established by occlusion of renal artery with artery forceps. The collagen concentration and the contents of collagen type Ⅰ, Ⅲ were measured. Meanwhile, the weight of heart and left ventricle was determined. Results (1) Both Tianma Gouteng Decoction and Captopril could reduce the left ventricle weight index (LV1) and collagen contents ( P 〈 0. 01 ) in RHR. (2) The results of scoring method indicated that the content of type I collagen obviously reduced in Tianma Gouteng Decoction group and in Captoril group as compared with that in the model group ( P 〈 0.05 or P 〈 0.01 ). Compared with Sham - operation group, the content of type Ⅲ collagen obviously reduced in model group and Captoril group ( P 〈 0.01) , but the difference was no significant between Tianma Gouteng Decoction group and Sham -operation group. Conclusion (1) Tianma Gouteng Decoction and Captopril can counteract myocardial fibrosis in RHR by reducing LVI and collagen content. (2) The mechanism of Captoprit in counteracting myocardial fibrosis is associated with the reduction of Type Ⅰ collagen content while that of Tianma Couteng Decoction is associated with the reduction of both Type Ⅰ and Type Ⅲ collagen contents.
