杏仁核毁损对甲基苯丙胺大鼠刻板行为和大鼠脑内5-HT_(2A)受体的影响 被引量:1

The effect of nucleus amygdalae lesion on 5-HT_(2A) receptor in brain in methamphetamine rats

摘要 目的探讨立体定向杏仁核毁损对甲基苯丙胺(MAP)大鼠脑内5羟色胺2A受体(5HT2A)表达的影响。方法24只SD大鼠随即分为对照组、模型组、假手术组和手术组,每组各6只;采用经腹腔注射MAP制备精神分裂症MAP模型,立体定向毁损杏仁核,聚合酶链反应技术观察5HT2A受体mRNA的表达。结果各组大鼠额叶、颞叶皮质和中脑均存在5HT2A受体mNRA(611bp)的阳性表达,且均以额叶皮质表达最为强烈;与对照组[(62.4±20.4)%]比较,模型组[(46.6±14.4)%]及假手术组[(48.5±15.2)%]大鼠中脑5HT2A受体mRNA受体表达的相对水平减低,差异有显著性(P<0.05);手术组[(57.2±18.4)%]5HT2A受体mRNA表达的相对水平高于模型组及假手术组,差异有显著性(P<0.05)。结论杏仁核毁损可能是通过使中脑5HT2A受体mRNA表达的增高而发挥治疗作用的。 Objective To explore the effect of nucleus amygdalae lesion using stereotactic technique on 5-HT2A receptor in brain in methamphetamine(MAP) rats. Methods 24 Sprague-Dawley rats were randomly divided into four groups:control group, MAP model group, pseudo-lesion group and lesion group. The nucleus amygdalae were destructed by radio-frequency (RF) using stereotactie technique, polymerase chain reaction (PCR) method was used to experiment the 5-HT2A receptor in brain in MAP rats. Results Expression of 5-HT2A receptor mRNA was present in the cortex of frontal lobe, cortex of temporal lobe and deutocerebrum . The relative expression levels of 5-HT2A receptor mRNA in deutocerebrum in MAP model group (46.6 ± 14.4) and pseudo-lesion group(48.5 ± 15.2) were significantly lower than that in control group(62.4 ± 20.4). But the levels of 5-HT2A receptor in deutocerebrum in lesion group (57.2 ± 18.4 ) was significantly higher than that in MAP model group and pseudo-lesion group. Conclusion Lesion of nucleus amygdalae could restrain the abaissemently expression of 5-HTaain deutocerebrum because methamphetamine employ in rats.
