摘要
目的:观察不同剂量枸杞多糖对D-半乳糖致衰老模型大鼠8-羟基鸟嘌呤糖苷酶表达的影响。方法:实验于2005-03/08在黑龙江省佳木斯大学生物化学教研室完成。①选用纯系Wistar大鼠40只。随机将大鼠分为4组:衰老组,枸杞多糖大剂量组,枸杞多糖中剂量组,枸杞多糖小剂量组,每组10只。衰老模型对照组:正常饮食,每日上午颈背部皮下按48mg/kg注射D-半乳糖,同时灌服温开水;枸杞多糖小剂量组、枸杞多糖中剂量组、枸杞多糖大剂量组:每组均正常饮食,除每日上午颈背部皮下按48mg/kg注射D-半乳糖外,同时分别按10,50,100mg/kg灌服枸杞多糖(市售枸杞多糖,用双蒸水分别制成10,50,100g/L溶液),连续给药45d。②用Trizol法提取肝组织核酸,反转录聚合酶链反应及凝胶成像技术检测8-羟基鸟嘌呤糖苷酶的mRNA表达水平。③计量资料差异比较采用方差分析和q检验。结果:大鼠40只均进入结果分析。大鼠肝脏组织的8-羟基鸟嘌呤糖苷酶mRNA表达水平:枸杞多糖小、中、大剂量组均明显低于衰老模型组(1.129±0.03,0.919±0.044,0.903±0.043,1.233±0.042,P<0.05),且随剂量的增加,呈下降趋势,枸杞多糖中剂量和大剂量组之间差异不明显。结论:枸杞多糖可以通过降低氧化损伤,减少DNA损伤,继而使修复酶8-羟基鸟嘌呤糖苷酶的表达下降而发挥抗衰老的作用。
AIM: To study the effect of lyeium barbarum polysaccharides at various dosages on expression of 8-oxoguanine in liver of aging model rats induced by D-galactose. METHODS: The experiment was completed in the Department of Biochemistry of College of Basic Medicine of Jiamusi University from March to August 2005. ① Forty pure-blood Wister rats were randomly divided into aging group, high, middle and low-dosage lycium barbarum polysacchrides groups with 10 in each group. Aging model control group: Rats were fed normally and were injected with 48 mg/kg D-galactose on the nap every morning, meanwhile, rats were perfused with warm boiled warter, Various dosage groups: Rats were fed normally and were injected with 48 mg/kg D galactose on the nap every morning, meanwhile, rats were perfused with 10, 50 and 100 mg/kg lycium barbarum polysaccharides for 45 days, Lycium barbarum polysaccharides in market were mixed with double-steamed water into 10, 50 and 100 g/L liquid respectively. ② Trizol reagent was applied to RNA extraction. Expressions of 8-oxoguanine mRNA were measured by reverse transcription polymerase chain reaction (RT-PCR) and gclatum imaging technique. ③ Measurement data were compared with analysis of wcriance and q test. RESULTS: All 40 rats were involved in the analysis of resuhs, Expressions of 8-oxoguanine mRNA in liver tissue: Expressions in various dosage groups were lower than those in aging model group (1.129±0.03, 0.919±0.044, 0.903±0.043, 1.233±0.042, P 〈 0.05). The expression was decreased With the increase of dosages. There was no significant difference between middle and high dosage groups. CONCLUTION: Lycium barbarum polysaccharides can decrease DNA damagc through inhibiting oxidative injury, and then delay aging through decreasing the expression of 8-oxoguaniue repairase.
