摘要
目的探讨隔山消对药物所致小鼠空间学习记忆障碍的作用,并初步探讨其作用机理。方法50只昆明小鼠随机平均分为空白对照组、模型组、阳性对照组(刺五加注射液)、隔山消高、低剂量组,各组动物在作相应的处理之后,于Mor-ris水迷宫实验中测试其潜伏期。各组动物完成行为实验后颈椎脱臼处死。在低温下取其脑组织,用羟胺比色法测定脑组织中Ach含量和Ach-E的活性。结果模型组动物达岸时间(即潜伏期)明显延长,表明空间学习记忆产生障碍;阳性对照组,隔山消高、低剂量组潜伏期均明显低于模型组,且第3天成绩好于空白对照组。三者相互比较,隔山消高剂量组第3天成绩最好。隔山消高,低剂量组的Ach含量明显高于模型组,而隔山消高低剂量组的AchE活性均略高于模型组,且均低于阳性对照组。结论隔山消对于药物所致的空间学习记忆障碍有一定的作用。
Objective To investigate the effects of Cynachum uilfordii (Maxim) Hemsl on the mouse space learning memory disorder caused by drug and to investigate the mechanism of effection. Methods 50 mouse were randomly divided into blank matched group, model group, positive control group( manyprickle acanthopanax root injection), Cynachum uilfordii high dosage group( 15 g/ kg) and low dosage group (7.5 g/kg). Each group was deal with severally, then measured the latent periods in Morris Water Maze test. The mouse of each group were put to death by dislocating their cervical vertebras after the behavior tests. The brain was hydroxylamine colorimetric method, Results The reaching-bank time ( latent period) of model group prolonged obviously, which showed the space leafing memory disorder;The latent periods of positive control group, Cynachum uilfordii high dosage group and low dosage group prolonged control group, Cynachum uilfordii high dosage group was the best of the three above. The content of Ach of Cynachum uilfordii high dosage group and low dosage group was visibly high dosage group and low dosage group were appreciably higher than that of model group,and both of them were lower than of positive control group. Conclusion The results indicated the Cynachum uilfordii( Maxim)Hemsl has effect on the space lcaring memory disorder caused by drug.
